نانوذرات مگنتیت (Fe3O4)، به دلیل خصوصیات منحصربه فرد فیزیکی، شیمیایی، حرارتی و مکانیکی دارای کاربردهای زیست پزشکی مختلفی مانند سلول درمانی، ترمیم بافت، تحویل دارو و تصویربرداری با رزونانس مغناطیسی اند. هدف از این مطالعه تولید نانوذرات مگنتیت با استفاده از میانکنش عصاره عاری از سلول جدایه های باکتریایی اندوفیت ریشه گیاه توت فرنگی و اکسید آهن فریک (Fe2O3) به عنوان پیش ساز نانوذرات بود.
مواد و روشهابرای سنتز برون سلولی نانوذرات مگنتیت، استراتژی عصاره عاری از سلول استفاده شد. ویژگی نانوذرات مگنتیت سنتزی پس از استخراج از مخلوط واکنش زیست تبدیلی، به وسیله آنالیزهای طیف سنجی مریی ماورای بنفش و الکترومیکروگراف های به دست آمده از میکروسکوپ الکترونی روبشی نشر میدانی، طیف سنجی پراش انرژی پرتو ایکس، آزمون پراش اشعه ایکس و طیف سنجی مادون قرمز فوریه مورد بررسی قرار گرفت.
نتایجبراساس نتایج به دست آمده، جدایه باکتری اندوفیت BR06 قادر به تبدیل زیستی پیش ساز اکسید آهن فریک به نانوذرات مگنتیت بود. تجزیه و تحلیل های فیلوژنیک تایید کرد جدایه باکتری مذکور متعلق به جنس باسیلوس است و ازنظر قرابت ژنتیکی بیشترین شباهت نوکلیوتیدی را با باسیلوس سیامنسیس (MT052693) دارد. سویه اندوفیت مذکور قادر به سنتز نانوذرات مگنتیت کروی به شکل گل کلمی با میانگین اندازه ذرات 34 نانومتری در غلظت 5 میلی مولار اکسید آهن فریک و پس از 24 ساعت واکنش بود. نتایج طیف سنجی و میکروسکوپ الکترونی سنتز نانوذرات مگنتیت را تایید کرد.
بحث و نتیجه گیریدر این پژوهش، برای نخستین بار زیست سنتز برون سلولی نانوذرات مگنتیت با استفاده از باکتری اندوفیت باسیلوس سیامنسیس تحت استراتژی عصاره عاری از سلول توصیف شده است.

تثبیت سویه های میکروبی دارای مزایایی مانند قابلیت تکرار و هزینه کمتر بازیافت سویه در فرایند تخمیر است. هدف از این مطالعه، بررسی اثر تثبیت سویه آیروموناس هیدروفیلا MSB16 بر توانایی تولید پروتیاز آن بود.
مواد و روشهابهترین روش تثبیت سویه MSB16، از میان 5 روش مختلف و با استفاده از پلیمرهای آلژینات، آلژینات - پلی وینیل الکل، آلژینات - پلی وینیل الکل - کربنات کلسیم، آلژینات - نانوذره گاما آلومینیوم اکسید و همچنین کربوکسی متیل سلولز انتخاب شد. مبنای انتخاب بهترین روش تثبیت، میزان تولید پروتیاز سویه تثبیت شده در مقایسه با سویه تثبیت نشده به صورت مطالعه یک فاکتور در یک زمان است. سپس فاکتورهای موثر در روش تثبیت انتخاب شده، با استفاده از روش شناسی سطح پاسخ، بهینه سازی شدند. آنالیز داده های مربوط به روش شناسی سطح پاسخ با استفاده از نرم افزار Design Expert 11 و آزمون تحلیل واریانس بررسی شد.
نتایجبراساس نتایج روش شناسی سطح پاسخ، بالاترین میزان تولید پروتیاز توسط سویه تثبیت شده، در پلیمر حاوی 3 درصد آلژینات - 3 درصد پلی وینیل الکل - 3 درصد کربنات کلسیم به دست آمد (U/mL 32/205). مدل درجه دوم به دست آمده، براساس مقدار F (59/25) و مقدار p (0001/0>) ازنظر آماری معنادار بود. بالابودن ضریب تشخیص (9584/0) و معنادارنبودن تست عدم برازش (7818/0) صحت مدل را تایید می کرد. تولید پروتیاز تحت تاثیر غلظت کربنات کلسیم و پلی وینیل الکل قرار می گرفت و با غلظت کربنات کلسیم رابطه معکوس داشت.
بحث و نتیجه گیریبهینه سازی آماری عوامل موثر بر تثبیت سویه MSB16 به روش آلژینات - پلی وینیل الکل - کربنات کلسیم به افزایش 13/2 برابری تولید پروتیاز نسبت به سلول های میکروبی آزاد و تثبیت نشده منجر شد.

اکتینومیست ها باکتری های رشته ای و گرم مثبت هستند که به صورت آزاد یا ساپروفیت و گاه همزیست با گیاهان دیده می شوند. این باکتری ها را می توان از همه اکوسیستم ها ازجمله خاک، آب، رسوبات دریایی به ویژه آب های گرم جداسازی کرد. اکتینوباکتری های دریایی به عنوان تولیدکننده طیف وسیعی از متابولیت های ثانویه شناخته شده اند. درواقع هر سویه از اکتینوباکتری ها به احتمال زیاد ظرفیت ژنتیکی برای تولید 10 تا 20 متابولیت ثانویه را دارد. حدود 23000 نوع آنتی بیوتیک از میکروارگانیسم ها کشف شده است که طبق برآوردها حدود 10000 نوع از آنها از اکتینوباکتری ها جدا شده اند. متابولیت های ثانویه تولیدشده توسط اکتینوباکتری های دریایی، طیف گسترده ای از فعالیت های زیستی مانند ضدباکتری، ضدقارچ، ضدسرطان، سمیت سلولی، سیتواستاتیک، ضدعفونی، ضدانگل، ضدمالاریا، ضدویروسی، آنتی اکسیدان و ضدرگ زایی و غیره دارند.
مواد و روشهادر این پژوهش برای جداسازی اکتینوباکتری ها، نمونه های آب و رسوب از خلیج فارس در محل ایستگاه انرژی اتمی بندر بوشهر جمع آوری شدند. برای کشت این نمونه ها از 7 نوع محیط کشت استفاده شد. برای تعیین هویت جدایه های به دست آمده، ویژگی های ریخت شناسی، تست های بیوشیمیایی و مطالعات مولکولی بررسی شدند.
نتایج24 جدایه اکتینوباکتر از نمونه های آب و رسوب سواحل خلیج فارس جدا شدند که 18 جدایه فعالیت ضدباکتریایی داشتند؛ بنابراین، 75 درصد جدایه ها مولد آنتی بیوتیک بودند. 95 درصد مواد به دست آمده از این 18 سویه، اثر ضدباکتریایی علیه باکتری های گرم مثبت و 60 درصد علیه باکتری های گرم منفی داشتند. 60 درصد از این مواد علیه هر دو گروه باکتری موثر بودند.
بحث و نتیجه گیرینتایج نشان دادند هرکدام از 18 نوع ماده ضدباکتری جداشده از 18 سویه مختلف، حداقل علیه یک باکتری بیماری زا موثر هستند که این نشان دهنده توانایی بالقوه این منبع برای مطالعه در زمینه دستیابی به آنتی بیوتیک های جدید است.

پوسته های زیستی که روی سطح خاک تشکیل می شوند، از میکرو و ماکرو ارگانیسم هایی همچون سیانوباکتری ها، قارچ ها، باکتری ها، اکتینومیست ها، جلبک ها، ویروس ها، خزه ها، گلسنگ ها و گیاهان تشکیل شده اند. سیانوباکتری ها، میکروارگانیسم های کلیدی در تشکیل پوسته های زیستی خاک، به خصوص در مناطق بیابانی اند که با رشد خود و تشکیل کلنی بر سطح خاک و تولید پلی ساکارید خارجی (EPS) باعث افزایش مقدار کربن و نیتروژن خاک می شوند و نقش مهمی در تقویت انواع خاک ها به ویژه خاک های فقیر از لحاظ مواد غذایی دارند و باعث جلوگیری از فرسایش آنها می شوند.
مواد و روشهادر این پژوهش، برترین سویه های سیانوباکتریایی مولد کربوهیدرات ترشحی، از میان 19 سویه جداشده از خاک استان های مختلف کشور انتخاب شد. میزان تولید کربوهیدرات در این سویه ها به روش فنول - اسید سولفوریک تعیین شد. سپس سویه های منتخب مولد بیشترین مقدار کربوهیدرات ترشحی، در مدل گلدانی به خاک های فقیر تلقیح شد. پس از گذشت 7 هفته از رشد سیانوباکتری ها در خاک، تغییر ویژگی های فیزیکی و شیمیایی آن اندازه گیری شد. همچنین سرعت تشکیل پوسته زیستی و میزان انسجام ذرات خاک به یکدیگر، به صورت میکروسکوپی بررسی شد.
نتایجمیزان تولید کربوهیدرات در این سویه ها به ترتیب برابر 66/86 میکروگرم بر میلی لیتر برای سویه Anabaena sphaerica، مقدار 91/55 میکروگرم بر میلی لیتر برای جنس Geitlerinema.sp و مقدار 19/64 میکروگرم بر میلی لیتر برای سویه Nostoc pruniformeبود. نتایج نشان می دهند هر سه این سویه ها قادر به تشکیل پوسته های زیستی در خاک هستند.
بحث و نتیجه گیریسویه Anabaena sphaericaبه دلیل سرعت رشد زیاد، تولید بیشتر زیست توده و کربوهیدرات ترشحی، افزایش بیشتر مقدار نیتروژن و کربن خاک و نیز تشکیل بهتر پوسته زیستی برای آزمون های بعدی انتخاب شد. این سویه برخی ویژگی های بافت خاک را بهبود بخشید که در حاصلخیزی آن می تواند تاثیر مثبتی داشته باشد.

روش های مختلفی برای تولید نانوذرات وجود دارند؛ اما امروزه سنتز نانوذرات با روش های زیستی به دلیل آسان بودن، سازگاری با محیط زیست و مقرون به صرفه بودن، توجه محققان را به خود جلب کرده است. هدف از پژوهش اخیر سنتز زیستی نانوذرات طلا با استفاده از اگزوپلی ساکارید لاکتوباسیلوس پاراکازیی (MN809528) و بررسی فعالیت ضدباکتریایی، ضدبیوفیلمی و آنتی اکسیدانی است.
مواد و روشهابرای سنتز نانوذرات طلا 30 میلی لیتر محلول 1 درصد اگزوپلی ساکارید (1/0 گرم) به حجم مساوی از محلول آبی HAuCl4 یک میلی مولار اضافه و با استفاده از استیرر (rpm 180) به مدت 48 ساعت در دمای اتاق به خوبی مخلوط شد. ویژگی نانوذرات طلا سنتزشده با استفاده از آنالیز های طیف سنجی UV-VIS، FT-IR، DLS، XRD، EDX، FE-SEM انجام شد. فعالیت ضدباکتریایی نانوذرات طلا تولیدشده با روش انتشار در چاهک، فعالیت ضدبیوفیلمی با روش رقت در میکروپلیت 96 خانه ای و فعالیت آنتی اکسیدانی نانوذرات با استفاده از قابلیت جذب رادیکال های DPPH بررسی شد.
نتایجداده های طیف سنجی، وجود پیک در 524 نانومتر را نشان دادند که ویژه نانوذرات طلا است. بررسی شکل و اندازه نانوذرات با FE-SEM نشان داد نانوذرات تولیدشده کروی شکل اند و به طور متوسط اندازه آن ها 50-20 نانومتر است. فعالیت ضدباکتریایی نانوذرات طلا با غلظت 50 میکروگرم بر میلی لیتر در برابر همه باکتری های تست مشاهده شد؛ با این حال، بیشترین فعالیت ضدباکتریایی در برابر اسینتوباکتر بومانی (ATCC 17978) مشاهده شد. در این مطالعه نانوذرات طلا تولیدشده به عنوان مهارکننده بیوفیلم و رادیکال DPPH عمل کردند.
بحث و نتیجه گیرینتایج نشان می دهند لاکتوباسیلوس پاراکازیی (MN809528)، منبع زیستی خوبی برای سنتز نانوذرات طلا است. مطالعه حاضر، نخستین گزارش از تولید نانوذرات طلا توسط اگزوپلی ساکارید جداشده از باکتری های اسید لاکتیک بومی است.

L- آسپاراژیناز یک آنزیم ضدسرطان مهم است که در صنایع پزشکی و غذایی کاربرد دارد. با در نظر داشتن اهمیت L- آسپاراژیناز در صنایع مختلف، یافتن سویه های مولد جدیدی ارجحیت می یابد که بتوانند سطح بالاتری از آنزیم با حداقل عوارض جانبی را تولید کنند.
مواد و روشهاغربالگری اولیه سویه های مولد L- آسپاراژیناز با آزمون کیفی روی پلیت محیط کشت زاپک داکس آگار تغییریافته حاوی L- آسپاراژین به عنوان منبع نیتروژن و فنل رد به عنوان اندیکاتور pH انجام شد. فعالیت L- آسپاراژینازی سویه مولد با استفاده از روش نسلریزاسیون اندازه گیری شد. شناسایی سویه قارچی با تعیین ویژگی های مورفولوژیک و آنالیز فیلوژنی ژن های ریبوزومی ITS (نواحی فاصله انداز رونویسی شونده داخلی) و ژن LSU (زیرواحد بزرگ ریبوزومی) انجام شد. تولید L- آسپاراژیناز با روش یک عامل در هر زمان بهینه سازی شد. تاثیر دما، میزان مایه تلقیح، منبع کربن و نیتروژن بر تولید آنزیم ارزیابی شد.
نتایجسویه Sarocladium kiliense IBRC-M 30453 از خاک، جداسازی و به عنوان یک سویه بالقوه مولد L- آسپاراژیناز شناسایی شد. روش تولید آنزیم خارج سلولی بود. فعالیت بهینه آنزیم U ml−1 4/62 در دمای 25 درجه سانتی گراد، میزان مایه تلقیح حجمی 9 درصد و با استفاده از سوکروز به عنوان منبع کربن و آمونیوم کلراید به عنوان منبع نیتروژن به دست آمد. فرایند بهینه سازی به افزایش 8/2 برابری تولید آنزیم منجر شد.
بحث و نتیجه گیریبه دلیل افزایش کاربردهای پزشکی و غذایی L- آسپاراژیناز در بازار تجاری، تقاضا برای این آنزیم از منابع جدید و با عوارض جانبی کمتر وجود دارد و L- آسپاراژیناز قارچی می تواند پاسخی مناسب به وضعیت کنونی بازار باشد؛ با وجود این، به کارگیری روش های آماری بهینه سازی و دست ورزی های ژنتیکی در مطالعات پیش رو برای دستیابی به حداکثر میزان تولید و بهبود قابلیت تولید صنعتی توصیه می شود.

Magnetic nanoparticles have various biomedical applications such as cell therapy, tissue repair, drug delivery, and magnetic resonance imaging (MRI) due to their unique physical, chemical, thermal, and mechanical characteristics as well as their suitable surface properties. The present study aimed to synthesize magnetic Fe3O4 nanoparticles (NPs) using the interaction of cell-free extracts of endophytic bacterial isolates with Fe2O3 as the precursor of NPs.
Materials and MethodsIn this study, the extracellular synthesis of Fe3O4 NPs was accomplished by the cell-free extract (CFE) strategy. Characterization of Fe3O4 NPs was performed using different methods, including ultraviolet–visible spectroscopy (UV–Vis), field emission scanning electron microscope (FE-SEM), energy-dispersive X-ray spectroscopy (EDX), X-ray powder diffraction (XRD), and Fourier transform infrared spectroscopy (FTIR).
ResultsThe obtained results of the study showed that the precursor Fe2O3 could be transformed into Fe3O4 NPs with the endophytic strain BR06. Phylogenetic analysis indicated that the isolated strainBR06 belonged to the genus Bacillus, and shared the highest sequence similarity with B. siamensis (MT052693). The Fe3O4 NPs with average diameters of 34 nm and cauliflower shape were synthesized by the endophytic strain BR06 in the biotransformation medium containing 5 mM Fe2O3 after 24 h incubation time. Electron microscopy and the associated spectroscopic analyses confirmed the formation of Fe3O4 NPs.
Discussion and ConclusionIn the present study, for the first time, the biosynthesis of extracellular magnetite NPs was described using B. siamensis under the cell-free extract strategy.

Microbial strain immobilization has some benefits like the repeatability and cost-effectiveness of strain recovery in the fermentation process. The present study aimed to investigate the effects of Aeromonas hydrophila MSB16 immobilization on its protease production.
Materials and MethodsThe best method for immobilizing MSB16 was selected using five different polymers including alginate, alginate-polyvinyl alcohol, alginate-polyvinyl alcohol-calcium carbonate, alginate-γ-Al2O3 nanoparticles, and carboxymethylcellulose. The best immobilization method was selected based on the ability of protease production of immobilized cells through the one-factor-at-a-time approach. Then, the selected method was optimized using the surface response methodology (RSM). Finally, the data were analyzed by Design Expert 11 software and the analysis of variance.
ResultsAccording to the results of RSM, the immobilized strain in a polymer containing 3% alginate - 3% polyvinyl alcohol - 3% calcium carbonate showed the highest protease production (205/32 U/mL). The second-order model was statistically significant based on the F-values (25.95) and p-values (0.0001). The high determination coefficient (0.9584) and the non-significance lack of fit (0.7818) confirmed the accuracy of the model. The MSB16 protease production was affected by calcium carbonate and polyvinyl alcohol concentrations and had an inverse relationship with calcium carbonate.
Discussion and ConclusionThe statistical optimization of the MSB16 immobilization method led to a 2.13-fold increase in its protease production than the free cells.

Actinomycetes are filamentous and gram-positive bacteria living as free-living, saprophyte, and sometimes in symbiosis with plants. These bacteria could be isolated from all ecosystems including soil, water, marine sediments and especially hot waters. Marine actinobacteria are known as the producers of a vast range of secondary metabolites. Any actinobacterial strain has the genetic potential for the production of 10-20 of the secondary metabolites. About 23000 antibiotic types were discovered from microorganisms which according to the estimations, about 10000 types have been isolated from actinobacteria. The secondary metabolites produced by marine actinobacteria have an extensive range of biological activities such as antibacterial, antifungal, anti-cancer, cytotoxic, cytostatic, ant-inflammatory, anti-parasite, anti-malaria, antiviral, anti-oxidant and anti-angiogenesis, etc.
Materials and MethodsIn the present study, water and sediment samples from the Persian Gulf at the Bushehr Atomic Energy Beach were collected for Actinobacteria isolation. For the cultivation of these samples, 7 culture medium types were used. For the identification of the gathered isolates, morphological properties, biochemical tests, and molecular studies (e.g. PCR) were investigated.
ResultsTwenty-four isolates of Actinobacteria were isolated from water and sediment samples of the Persian Gulf coast. Eighteen of the isolates have antibacterial activities. Seventy-five persent of the isolates were antibiotic producers and 95% of the gathered substances from these 18 strains had an antibacterial effect against gram-positive bacteria and 60% against gram-negative bacteria. Also, 60% of these substances were effective against both bacteria groups.
Discussion and ConclusionThe abundance of marine actinobacteria in new sources of the Persian Gulf and the extension of this sea’s coat indicates the potential capability of this source for investigation in the scope of attaining new antibiotics.

Biological soil crusts including cyanobacteria, fungi, bacteria, actinomycetes, algae, viruses, mosses, lichens, and plants are formed on the soil surface. Cyanobacteria are key microorganisms in the formation of the biological soil crust, especially in arid and desert areas. Cyanobacteria increase carbon and nitrogen content in soil via increasing colony formation and Exopolysaccharide (EPS) production and play an important role to strengthen soil structure especially in poor soil and prevent it from eroding.
Materials and MethodsIn the present study, three cyanobacteria with the highest secreted carbohydrate production were selected among 19 isolates of different provinces’ soil. The amount of carbohydrate production of the selected strains was determined by using the phenolic sulfuric acid method. Then, three selected strains, with the highest amount of secreted carbohydrate were inoculated to the pot filling with poor soils. After 7 weeks of cyanobacterial inoculation, some physical and chemical alterations of the soil were measured. Furthermore, the rate of formation of the biological soil crust and the adhesiveness of the soil particles were microscopically investigated.
ResultsThe carbohydrate content was 86.66 µg/ml for Anabaena sphaerica, 55. 91 µg/ml for Geitlerinema.sp, and 64.19 µg/ml, for Nostoc pruniforme. The results of the study also show that these three strains are capable of forming biological soil crust on the soil surface.
Discussion and ConclusionAnabaena sphaerica, due to its high growth rate, more biomass production, and more secreted carbohydrates increased soil nitrogen and carbon content, as well as better formation of biological soil crust was selected for further investigation. This strain improved some soil texture characteristics and probably it can have a positive effect on soil fertility.

Although there are different methods for the production of nanoparticles, today the synthesis of nanoparticles with biological methods has attracted the attention of researchers because it is an easy, environmentally friendly, and cost-effective method. The present study investigates the biosynthesis of gold nanoparticles using Lactobacillus paracasei (MN809528) exopolysaccharide and studies the antibacterial, anti-biofilm, and antioxidant properties.
Materials and MethodsFor the synthesis of gold nanoparticles, 30 ml of 1% exopolysaccharide (0.1 g) solution was added to an equal volume of 1 mM aqueous HAuCl4 solution and mixed well using stirred (180 rpm) for 48 h at room temperature. The characterization of synthesized gold nanoparticles was performed using UV-VIS, FT-IR, DLS, XRD, EDX, FE-SEM spectroscopic analysis. Antibacterial activity of produced gold nanoparticles by agar well diffusion method, anti-biofilm activity by 96 well microplate dilution method, and antioxidant activity of nanoparticles using DPPH radical adsorption capability were examined.
ResultsSpectroscopic data showed the presence of a peak at 524 nm, which is specific to gold nanoparticles. Examination of the shape and size of nanoparticles with FE-SEM showed that the produced nanoparticles were spherical in shape and their average size is 2o-50 nm. Antibacterial activity of gold nanoparticles at a concentration of 50 µg /ml was observed against all bacterial tests. However, the highest antibacterial activity was observed against Acinetobacter baumannii (ATCC 17978). In the present study, the gold nanoparticles produced acted as biofilm and DPPH radical inhibitors.
Discussion and ConclusionThe results of the study show that Lactobacillus paracasei (MN809528) is a good biological source for the synthesis of gold nanoparticles. The study is the first report on the production of gold nanoparticles by exopolysaccharides isolated from indigenous lactic acid bacteria.

L-asparaginase is a significant anticancer enzyme used both in medicine and food industries. Considering the importance of L-asparaginase in different industries, finding new sources of producer strains that can produce higher levels of this enzyme with minimum side effects is preferred.
Materials and MethodsInitial screening for L-asparaginase-producing strain was performed via qualitative plate assay on modified Czapex Dox's agar medium using L-asparagine as the nitrogen source and phenol red as pH indicator. L-asparaginase activity of the fungal strain was quantified by the nesslerization method. Its identification was performed by using both morphological characteristics and phylogenetic analyses of DNA sequence data, including ribosomal DNA regions of ITS (Internal Transcribed Spacer) and LSU (large Sub-Unit) rDNA. L-asparaginase production was optimized by the One Factor-At-the-Time (OFAT) technique. The impacts of temperature, inoculum size, nitrogen, and carbon sources on the enzyme production were then evaluated.
ResultsSarocladium kiliense IBRC-M 30453 was isolated from soil and identified as a potent enzyme-producing strain. The enzyme production was based on the extracellular mode. An optimum enzyme activity of 62.4 U ml−1 was obtained at the temperature of 25°C with inoculum size of 9% (v/v) and sucrose containing carbon and ammonium chloride as the nitrogen source. The optimization process led to 2.8-fold increase in the enzyme production.
Discussion and ConclusionThere is a market demand for an alternate source of L-asparaginase with fewer adverse effects due to an increase in the clinical and industrial applications of this enzyme. Fungal L-asparaginase can be the proper answer to the current status of the market. However, application of statistical optimization processes and genetic manipulation have been recommended in other studies to achieve its maximum production level and improve feasibility of its industrial production.
